Abstract

|
The properties and functions of mistletoe lectin have been studied for many years in European countires for its immune enhancing and anticancer activities. Recently, the studies to unravel the function of Korean mistletoe lectin are vigorously persued. In the previous studies, we have demonstrated that Korean mistletoe extract has immune enhancing and cytokine inducing activities. It has been reported that these activities of Korean mistletoe are due to the lectin components which are composed of disulfide-bonded two subchains. Here, we purified an isoform (KML-IIU) and its subchains (A and B) from Koeran mistletoe lectin by affinity chromatography using a£Þ-mercaptoethanol, and examined their activity to induce TNF-a¢¥ in RAW 264.7 murine macrophage cells using ELISA kit. We also investigated their TNF-a¢¥ inducing activities in aspect of mRNA expression by Northern blot analysis. The data show that KML-IIU and its A and B subchains induce TNF-a¢¥ production from RAW 264.7 cells, and TNF-a¢¥ inducing ability of B chain was 20-fold higher than A chain.
